چکیده

The discovery that the absorption bands at 293, 281-5 and 271 m,u. (Heilbron, Kamm & Morton, 1926, 1927) present in fractions obtained in the recrystallization of cholesterol were shown very strongly by ergosterol (Rosenheim & Webster, 1927) focused interest on the latter and culminated in the isolation ofcalciferol (vitamin D2). Some time later it was recognized that 7-dehydrocholesterol was a provitamin D and vitamin D3 was eventually obtained pure. Vitamin D. in fact appeared to be the principal, if not the only 'natural', vitamin D. With the achievement of commercial production interest in the field declined, although many problems remained unsettled. Perhaps the most important of these is the biogenesis ofprovitamin D. In the herbivorous animal, the dietary sterols, sitosterol, stigmasterol, etc., are unlikely to be responsible for the appearance of 7dehydrocholesterol. Ergosterol, which is characteristic of yeasts and fungi, could scarcely account for the wide distribution of provitamin D3 in animals. The present work provides evidence ofa reversible enzymic dehydrogenation of cholesterol mainly in the gut wall. The 7-dehydrocholesterol is thus not of dietary origin. It may have biological functions independent of its role as provitamin D3.

The "d" Vitamins

In a classical sense, vitamin D3, the form produced in animals, is not a true “vitamin” because it is produced in the skin from 7-dehydrocholesterol by UV radiation in the range 290 to 300 nm; 7-dehydrocholesterol is produced from cholesterol metabolism. Only when exposure to sunlight is inadequate does vitamin D3 become a vitamin in the historical sense. Further, vitamin D3 is now termed a pro...

Sterol metabolism. 6. The interconversion of cholesterol, 7-dehydrocholesterol and lathosterol in the rat.

The three closely related sterols, cholesterol, 7-dehydrocholesterol and lathosterol, occur in animal tissues in different proportions. From their structures they would appear to be interconvertible by simple dehydrogenation or reduction steps as shown in Fig. 1, but unequivocal evidence for such a mechanism has not yet been obtained. It has generally been assumed that 7-dehydrocholesterol (pro...

Isolation and identification of 24-dehydroprovitamin D3 and its photolysis to 24-dehydroprevitamin D3 in mammalian skin.

Until now it had been assumed that mammalian skin contains only one provitamin D, 7-dehydrocholesterol, that is eventually converted to vitamin D3 after the skin is exposed to sunlight. Examination by reverse phase high performance liquid chromatography of lipid extracts from young rat skin, however, led to the observation that 7-dehydrocholesterol is not the only provitamin D in rat skin. Anot...

Effect of a new inhibitor of cholesterol biosynthesis (AY 9944) on serum and tissue sterols in the rat.

AY 9944 is a novel inhibitor of cholesterol biosynthesis which appears to act on the Delta 7-reductase enzyme, interfering with the conversion of 7-dehydrocholesterol to cholesterol. In all tissues studied, with the exception of the lens, there was a reduction in cholesterol and a marked accumulation of 7-dehydrocholesterol. Isotope studies showed a reduced uptake of acetate into cholesterol, a...

A novel pathway for hormonally active calcitriol.

Calcitriol [1alpha,25(OH)2D3], the hormonally active form of vitamin D3 (D3) is produced in both renal and extrarenal tissues. Our findings demonstrate that physiological doses of UVB radiation at 300 nm induce the conversion of 7-dehydrocholesterol (7-DHC) via preD3 and D3 into calcitriol in the pmol range in epidermal keratinocytes. The hydroxylation of photosynthesized D3 to calcitriol is st...
